脑中小胶质细胞诱导的炎症反应在缺血/再灌注(I/R)脑损伤中起关键作用。已有研究表明,小胶质细胞激活与TLR4/NF-κB信号通路密切相关。注射丹酚酸(SAFI)已被用于临床实践中以治疗缺血性中风,具有报道的神经保护作用,但是其潜在的机制仍然不确定。
2016年11月,天津中医药大学研究团队,就注射丹酚酸治疗缺血性中风的机制进行了一系列实验,并将相关结果发表在期刊《Journal of Ethnopharmacology》。
首先,实验员研究了SAFI对内毒素刺激的BV-2小胶质细胞炎症反应的影响。并进一步采用大脑中动脉闭塞大鼠模型,探讨SAFI治疗缺血性卒中的机制。结果显示,与对照组相比,SAFI治疗显著降低了神经炎症和梗死体积。SAFI处理后缺血半球小胶质细胞激活减少,TLR4 /NF-κB信号被明显抑制,细胞因子IL-1β和IL-6表达和释放减少。

这项研究提供了SAFI有效保护脑缺血后大脑的证据,这可能是通过减轻小胶质细胞的炎症而引起的。目前的结果提供了强有力的实验证据,支持SAFI作为至少部分通过TLR4-NF-κB炎症信号传导途径治疗脑缺血/再灌注损伤中小胶质细胞介导的炎症的治疗策略。

The results of the present study suggest that SAFI can protect cerebral ischemia-reperfusion injury in rats and suppress inflammatory responses by activated microglia in vitro. SAFI attenuates NF-κB and TLR4 expression and inhibits the nuclear translocation of NF-κB, decreasing mRNA expression involved in the TLR4-NF-κB signaling pathway. This suggests that NF-κB plays a critical role in SAFI-mediated neuroprotective effects. Notwithstanding, the present results have provided strong experimental evidence supporting SAFI as a therapeutic strategy to treat microglia-mediated inflammation in cerebral ischemic/reperfusion injury at least partly by the TLR4-NF-κB inflammatory signaling pathway.
